Agricell Report about a paper that was presented at the Society for In Vitro
Biology's World Congress on In Vitro Biology, held 10-15 June 2000 in San
Diego, California.

In Vitro Collection of Leaf and Bud Tissues

At a workshop on "the challenge of in vitro collection and field collections
for tissue culture," V. Pence of the Cincinnati Zoo and Botanical Garden
described an in vitro method that her group has used to collect leaf and bud
tissues from Trinidad, Costa Rica, Cincinnati, North Carolina, Florida and
Utah. Leaf or bud tissues are removed from plants, surface disinfested with
70% ethanol, and transferred to small vials containing sterile, semisolid
Linsmaier-Skoog basal medium supplemented with 3% sucrose, 0.22% phytagel,
and 0.5 mg/L each of BAP and NAA. To control fungal contamination, active
benlate is added to the medium at a level of 100 mg/liter. Bacterial
contamination is controlled by adding a drop of antibiotic solution to the
top of the explant or by adding plant preservative mixture (PPM) to the
medium at a concentration of 0.2-0.25 ml/liter.
    With the use of this field collecting technique, Pence's group has
obtained 30-70% of  cultures free of visible contamination, depending on the
collection site. In general, contamination was higher in cultures obtained at
tropical locations as compared with those collected in temperate regions.
    Pence stated, "Continued work should improve the technique, but even
current methods of in vitro collection offer a significant opportunity for
the collection of plant germplasm when seeds are not available."
