   The attachment is one of my construction plans.
   I want to use the binary vector pYF133 using my cDNA instead of GFP gene in the vector. 
As for the four NotI sites on the pYF133 and without proter sites blanking my cDNA in pGEM-T 
Easy cloning vector, I designed that plan for my construction.
