Re: Problem with zeatin stock preparation

Dear Diskus,
If your Zeatin is from Sigma, then three dissolve well in 1N NaOH. But one
do not go well in NaOH. You can add HCl in it, this is not going to harm
you culture or medium. However, you will need to checkout the pH of
medium, how much it change i.e. prepare 100 ml liquid medium, set pH
5.8...., Autoclave, and add Zn and see the fianl pH. If goes down like 5.4
then set initial pH 6.0. since in general pH of meidum after autoclave is 5.6.
good luck

 Dear all,

 I have a question regarding zeatin stock preparation. I prepared a 10 mg/ml
stock and to dissolve zeatin i used 1M NaOH. Maybe i used to much NaOH and
after freezing, the solution was purple. If i add some HCl it turns
transparent again, but i don't know if i still can use that zeatin. Any help
will be very welcome.

 Thanks in advance,

 Antonio
